วัตถุประสงค์
:
2.1 To perform a heterologous gene expression in KJ134 by constructing the plasmid containing sucrose-utilizing genes derived from E. coli B, which is able to utilize sucrose naturally.
2.2 To study the effect of sucrose and cane molasses on the production of succinate in titer, yield, and productivities in KJ134 harboring the recombinant plasmid.
2.3 To study the carbon flux distribution from sucrose into intracellular metabolite intermediates. The insights information from metabolic flux analysis would lead the further strain improvements to obtain an efficient succinate-producing strain, which is promising an industrial need for exhibiting the high production yield, titer, and productivities of succinate from sucrose and molasses. Moreover, this research achievement will be a platform to establish further strain development for utilizing another carbon sources such as starch and cellulosic substrates to produce succinate.
