วัตถุประสงค์
:
1. Evaluation of the in vivo putative AR-antagonistic and/or ERs-agonistic effects of EQ on brain function, particularly the neuroendocrine regulation of the intact LHPT axis, and pituitary function by measuring the levels of mRNA and protein expression of genes implicated in the neuroendocrine regulation of the LHPT axis in the brain (MBH/ME, MPOA/AH, amygdala, striatum, hippocampus) and pituitary in the adult male rat after 5 days oral treatment. As the reference compounds, the pure AR-antagonist, flutamide, and pure ERs-agonist, estradiol, will be employed.
2. The possibility of the short-term (5 days) in vivo oral administration in adult male rats utilizing both the classical endpoints (reproductive organ weights, serum reproductive hormone levels) suggested by the OECD and the additional endpoints demonstrated in this study (gene expression levels) as an accurate, rapid, reliable, and cost effective in vivo screening assay for assessing the in vivo putative endocrine-active chemical and environmental compounds with regard to neuroendocrine and reproductive aspects.
